I was wondering if i spend less than $50 after coupon, could i still use this coupon to get extra points? or it dpends on the cashier?

Should be able to use that.

I'd make sure the cashier scans it and then hand them the coupons.

Won't make a difference in their system about the order but the cashier may refuse to scan it if they see you are using coupons and notice you drop below the $50.
Just make sure none are FPC and they are rung in under 2001
